Visão Geral

A mitocôndria é um organelo crítico no contexto da fisiologia celular. Funciona como central energética da célula, produzindo a maioria da energia química, desempenhando ainda papeis muito importantes na regulação da morte e controlo de qualidade celulares. Por serem participantes ativos na homeostasia redox e no controlo do cálcio celular, o nosso grupo tenta fornecer avanços significativos no conhecimento das alterações mitocondriais contextualizadas no metabolismo, sinalização redox e respostas de stress em toxicidade química, cancro, doenças cardiovasculares e hepáticas, envelhecimento e diferenciação de células estaminais.

O nosso foco é determinar se a regulação intrínseca, farmacológica ou não farmacológica (exemplo: mediada pelo exercício físico) da biogénese, metabolismo e controlo de qualidade mitocondrial é capaz de reduzir a lesão do órgão durante diferentes patologias ou causada pela toxicidade de xenobióticos. Mecanismos de biogénese mitocondrial e sua regulação por moléculas como o resveratrol ou proteínas como sirtuinas têm sido importantes questões experimentais. Outro objetivo importante, é investigar de que forma a actividade mitocondrial regula a diferenciação de células estaminais (cancerígenas).

Diferentes moléculas ou patologias activam vias de toxicidade/stress mitocondriais, afetando o fenótipo celular e frequentemente a sobrevivência do órgão. Entre os diferentes tóxicos mitocondriais, destacamos agentes cancerígenos como antraciclinas, anti-estrogénios ou retinoides. Desenvolvemos uma biblioteca de moléculas dirigidas à mitocôndria que poderão ser promissores agentes anti-cancerígenos ou antioxidantes.
Considerando as alterações mitocondriais durante o processo de envelhecimento, investigamos o papel da disfunção bioenergética mitocondrial e do desarranjo da beta-oxidação de ácidos gordos na mitocôndria e peroxisomas na menopausa associada ao decaimento de estrogénios.
